Riley was awarded the Reggae Runnins Caribbean Heritage Award at the 2009 HOT 105 FM Caribbean-American Heritage Awards. His commercial breakthrough came in 2006 with the Parables album and the Jamaican number one single "She's Royal". Contagious features guest appearances from Damian Marley, Vybz Kartel, Etana, Konshens, Demarco and Duane Stephenson. Riley has released three albums, Challenges (2004), Parables (2006), and Contagious (2009). He was guided in his early career by Dean Fraser, who also performs in his band and produces much of Riley's work. He began his musical career as a deejay working with Busta Rhymes, taking his birth sign as his stage name at his mother's suggestion (the spelling later changed from Taurus to Tarrus), and recorded his first singles for his father's Love & Promotion label. He was born in the Bronx and grew up in South Florida. Riley is the son of veteran singer Jimmy Riley and nurse Lavern Tatham-Riley. Except for the same surname, Tarrus, while clearly not possessing his famous father's voice with its sharp edge, is content musically to create his own legacy. His songwriting is bold, yet definitive, passionate and authentic, without being strident or overly militant. It conveys his every mood, concern and context eloquently. John’s Background Switcher (or JBS for short) periodically changes the background image on your Windows computer to something awesome (if you’re a Mac user, go here instead ). Also, the app loses most of its best features if you lose internet connectivity. The first thing is that the app runs constantly in the background, and uses a marginal amount of system resources when pulling new wallpapers. While John’s Background Switcher is effective at its job and feature rich, it isn’t without a couple drawbacks. Shut off when a certain application is detected to be running. The program has nearly every setting, feature, and option that you could possibly need for handling Windows desktop backgrounds: Most notable about JBS is the advanced settings. It can also save a particular photo if you’re viewing a stream of one you fancy. The system tray icon for JBS offers a lot of settings, with the most useful one being to rotate to a new background. It’s certainly not a bad way to sync wallpapers across multiple machines that might not always be on the same local network. It does this by authorizing through your online account, and then it can display photos that are not visible to the public. The app can also be designed to display files from your personal collection on Flickr, Facebook, Instagram, Vladstudio, and Picasa. It doesn’t just work with online sources, it can also pull images from folders on your computer.Īnother groovy feature, if you have a dual monitor setup, is displaying different pictures on each monitor. The rear section of the PG-1 consisted of a central tube holding the propelling charge, and a second tube around this carrying the fins. The design of the PG-2 differed considerably from that of the PG-1 of the RPG-1. The larger cartridge gave the PG-2 warhead slightly better practical range as well, about 150 m (490 ft) against stationary targets. This improved penetration to 180 mm (7.1 in), which allowed it to penetrate the frontal armor of all but the very heaviest tanks, and the side and rear armor of any tank. The RPG-2 used a custom designed 40 mm (1.6 in) cartridge to provide much greater power, and the warhead enlarged to 80 mm (3.1 in). The main difference in performance between the two were due to size. Development of the RPG-2 was carried out by the GSKB-30 design bureau, originally part of the Commissariat for Munitions, but in the post-war period handed to the Ministry of Agriculture to help design farm equipment. Modifications to improve this began, but in 1947 the RPG-2 program began as a parallel project. The warhead was already straining the abilities of the cartridge and its range was already considered too low. Įarly testing revealed several minor problems, but, by the time these were being solved, 150 mm of penetration was no longer considered effective against modern tanks, even late-war designs like the Panther. Propelled by a 30 mm cartridge, the 70 mm (2.8 in) high-explosive anti-tank (HEAT) shaped charge round could penetrate about 150 mm (5.9 in) of homogeneous armour. Studying German and US anti-tank rocket designs, in 1944 the Soviets began development of the RPG-1 with the goal of combining the best features of the German Panzerfaust single shot recoilless weapon with the US Bazooka rocket launcher. The basic design and layout was further upgraded to produce the ubiquitous RPG-7. The RPG-2 offered better range and armor penetration, making it useful against late and post-World War II tanks, in contrast to the RPG-1 that had only marginal utility. It was the first successful anti-tank weapon of its type, being a successor to the earlier and unsuccessful rocket-propelled grenade RPG-1. The RPG-2 ( Russian: РПГ-2, Ручной противотанковый гранатомёт, Ruchnoy Protivotankovy Granatomyot English: "hand-held antitank grenade launcher") is a man-portable, shoulder-fired anti-tank weapon that was designed in the Soviet Union. In biological systems, the synthesis of nucleic acids, such as DNA and RNA, is catalyzed by enzymes in various aqueous solutions. Our engine can be used to profile Ψ-occupancy across cell types and cell states, thus providing critical insights about physiological relevance of Ψ modification to mRNAs. We show that applying our model is critical for quantification, especially in low-abundance mRNAs. Our supervised machine learning models reveal that for every site studied, different signal parameters are required to maximize Ψ classification accuracy. In this work, we apply supervised machine learning models that are trained on sequence-specific, synthetic controls to endogenous transcriptome data and achieve the first quantitative Ψ occupancy measurement in human mRNAs. Nanopores may be used to directly identify Ψ sites in RNAs using a systematically miscalled base, however, this approach is not quantitative and highly sequence dependent. The pseudouridine (Ψ) mRNA modification is also highly abundant but difficult to detect and quantify because there is no available antibody, it is mass silent, and maintains canonical basepairing with adenine. Existing studies have typically focused on the identification and impact of adenine modifications on mRNA (m ⁶ A and inosine) due to the availability of analytical methods. Our proof-of-concept experiments for higher-fidelity incorporation of uridine analogs during IVT provide guidelines when choosing RNAPs for the generation of modified uridine-containing mRNAs in vitro.Įnzyme-mediated chemical modifications to mRNA are important for fine-tuning gene expression, but they are challenging to quantify due to low copy number and limited tools for accurate detection. Based on our findings, we introduce a novel method to improve uridine analog incorporation fidelity during IVT. We also show that the array of nucleotide misincorporation is not dependent on the template DNA sequence context and that the distribution of these misincorporated nucleotides is not localized to any specific region along the length of the RNA. The fidelity of nucleotide incorporation differs between RNA polymerases however, the spectrum of mutations observed between the RNAPs is similar. We demonstrate that m1Ψ is incorporated with higher fidelity than Ψ. To decipher the fidelity with which these modifications are incorporated during the in vitro transcription (IVT) process, we compared the incorporation fidelity of uridine analogs with different RNA polymerases. To overcome the inherent immunogenicity, as well as to increase the therapeutic efficacy of the molecules, uridine analogs-such as pseudouridine (Ψ) and N1-methyl-pseudouridine (m1Ψ), are incorporated in the synthetic mRNA. In vitro transcribed synthetic messenger RNAs (mRNAs) represent a novel therapeutic modality. These results may aid in future efforts that employ RNA polymerases to make therapeutic mRNAs with sub-stoichiometric amounts of m1Ψ. The SP6 polymerase introduced m1ΨTP when competed with UTP with a smaller window of yields (15-30%) across all sequence contexts studied. Experiments with SP6 RNA polymerase, as well as chemically-modified triphosphates and DNA templates provide insight to explain the observations. A significant sequence context dependency was observed for T7 RNA polymerase with insertion yields for ΨTP versus UTP spanning a range of 20-65%, and m1ΨTP versus UTP producing variable yields that differ by 15-70%. Understanding the nanopore signatures for Ψ and m1Ψ enabled a running start T7 RNA polymerase assay to study the selection of UTP versus ΨTP or m1ΨTP competing mixtures in all possible adjacent sequence contexts. The base calling data for Ψ or m1Ψ were similar but different from U allowing their detection. Direct RNA sequencing with a commercial nanopore platform was used to sequence RNA containing uridine (U), pseudouridine (Ψ) or N1-methylpseudouridine (m1Ψ) in >100 different 5-nucleotide contexts.
